Anti-viral Activity of Recombinant Human Alpha Interferon Expressed on Saccharomyces cerevisiae

Park Soon-Jae
Abstract
The cell sensitivity of recombinant human alpha interferon(LBD-007) expressed in Saccharomyces cerevisiae was studied in bovine (MDBK, EBTr), human (FL, HEp-2,WISH, A549, HeLa, HeLa S3), and mouse (L929) cells. For the systematic experiments, cytophathic effect / dyeup take method was employed. The results showed that human cells were most sensitive to the anti-viral effect of alpha interferon when challenged by Encephalomyo carditis virus (EMCV). On the contrary, the antiviral activity of alpha interferon against Vesicular stomatitis virus (VSV) was highest with the bovine cells. When VSV was used as a challenge virus, MDBK cell was most appropriate with regard to the sensitivity and reproducibility of bioassay. On the other hand, WISH cell gave the best result by use of EMCV.
